A side quest from Cosmo Canyon has you measuring Mako Frequency at certain locations. This From Whence Life Flows Guide For Final Fantasy 7 Rebirth will tell you where you can find each of the special Lifesprings that Bugenhagan asks you to find, as the normal Lifespring locations are not used in this quest.
Lifespring Location #1
You can find the first Lifespring at the location shown above. It’s just South of Cosmo Canyon. You can simply approach through the desert, you don’t need to worry about any difficult to locate path or flying sections. Simply head straight to the location and interact with the wooden gate. Bugenhagan will appear and hand you a key. You can then use that to open the gate and continue with the quest.
Lifespring Location #2
You can find the second Lifespring at this location. As before, it’s a simple approach from ground level in the desert. There is a small group of enemies that are attacking the door, you need to defeat them before you’re able to open it. Head inside to complete this objective.
Lifespring Location #3
You can find this Lifespring at this location, a short distance from the previous Lifespring. There’s a long road that leads up the side of the cliff. Follow it up part of the way and a group of ruins will be on your left. Search the top of those ruins to find the door to the Lifespring.
Lifespring Location #4
The next Lifespring can be found in the central part of the Cosmo Canyon Region, just South West of Cosmo Canyon itself. There are another group of enemies at this gate, so make sure to defeat them before trying to get inside.
Lifespring Location #5
You can find the final Lifespring at this location, a short walk from the nearby Chocobo Stop.
